If transmission what?
Some Toyotas of that age had problematic transmissions, Toyota extended the warranty. Check with your dealership.

The OP might be asking if using the paddle shifter hurts the transmission.
Who knows, we may never know…

As a 2014 Toyota Highlander owner I’m also a member of a Highlander forum. The Lexus RX-350 is basically the same vehicle. Starting in 2017 with the introduction of the 8-speed transmission Toyota has had a slew of transmission failures. There have been a variety of class-action lawsuits already.
